目的探讨钙结合蛋白S100A12在小鼠重症急性胰腺炎治疗中的潜在作用。方法采用连续7次腹腔注射雨蛙素50μg/kg(每次间隔1 h)及脂多糖建立小鼠急性胰腺炎模型。将160只SPF级雌性ICR小鼠随机分为对照组(A组,生理盐水)、轻型组(B组,雨蛙素)、重症组(C组,雨蛙素+脂多糖)、干预组(D组,S100A12重组抗体+雨蛙素+脂多糖),每组40只。在首次注射雨蛙素后8 h、12 h和24 h采血,分别检测各时段血清中S100A12、淀粉酶(AMY)、C反应蛋白(CRP)、白介素(IL-1β、IL-6)、肿瘤坏死因子(TNF-α)的含量,观察小鼠胰腺的病理形态并进行评分。结果与C组相比,D组各时段血清中AMY、CRP、IL-1β、IL-6、TNF-α、S100A12含量明显降低(P0.05);与B组相比,D组各时段血清S100A12浓度明显降低(P0.05),胰腺病理形态也有明显改善。结论 S100A12重组抗体能够有效降低小鼠急性胰腺炎的严重程度,S100A12可以作为重症急性胰腺炎治疗的一个有效靶点。  相似文献

In the first three days after admission cranial ultrasound was performed in each child. Results The difference among the L??H and N group was statistically significant ??P2=17.20??P??0.05??; S100B and GFAP levels in L and H group were positively correlated ??γ= 0.658?? 目的 研究脑脊液中新蝶呤(NPT)、S100b水平在诊断儿童中枢神经系统感染中的价值。方法 采用ELISA法测定中枢神经系统感染患儿脑脊液NPT及S100b浓度,对比感染及非感染患儿之间指标的异同,并采用ROC法评价两种指标的诊断价值。结果 病毒性脑炎患儿脑脊液中NPT及S100b浓度较对照组及化脓性脑膜炎患儿明显升高,差异有统计学意义(PP>0.05),但化脓性脑膜炎患儿S100b水平明显高于对照组(P结论 NPT及S100b在儿童中枢神经系统感染的诊断中有一定的价值,且在诊断病毒性脑炎中作用明显,尤以S100b指标为佳。  相似文献

目的 探讨常规栓塞化疗 (TOCE)与节段性栓塞化疗 (S TOCE)在小肝癌治疗中的疗效比较。方法  5 6例中2 6例行TOCE治疗 42次 ,平均 1.6次 /例 ,3 0例行S TOCE治疗 3 9次 ,平均 1.3次 /例。术后随访 6~ 3 6个月以上 ,并行B超、螺旋CT(SCT)及DSA (再次治疗时 )检查 ,以判断疗效。结果  2 6例行TOCE治疗病例 ,碘油完全充填 12例 ,占46.2 % ,-年内复发 5例 ,占 19.2 % ;3 0例行S TOCE治疗病人 ,碘油完全充填 2 5例 ,占 83 .3 % ,一年内复发 2例占 6.7% ;TOCE与S TOCE在小肝癌治疗中完全充填及术后复发等比较有显著差异 (P <0 .0 1)。结论 S TOCE是小肝癌非手术治疗首选方法。在疗效上与TOCE比较有显著差异  相似文献

OBJECTIVES: The aims of this study were to characterize the microflora in crevices around titanium orthodontic anchor plates using anaerobic culture and molecular biological techniques for bacterial identification, and to compare the microbial composition between crevices around anchor plates and gingival crevices. MATERIAL AND METHODS: Samples from crevices around titanium anchor plates and healthy gingival crevices of 17 subjects (aged 20-29) were cultured anaerobically, and isolated bacteria were identified by 16S rRNA sequencing. RESULTS: The average logarithm colony-forming units/ml were 6.84, 7.51 and 8.88 in healthy anchor plate crevices, inflamed anchor plate crevices and healthy gingival crevices, respectively, indicating that the bacterial density of anchor plate crevices was lower than that of healthy gingival crevices. Of 184 strains isolated from healthy anchor plate crevices of seven subjects, 108 (59%) were anaerobic bacteria, while 73 (40%) were facultative bacteria. Predominant isolates were Gram-negative rods, such as Campylobacter (12%), Fusobacterium (10%) and Selenomonas (10%), and Gram-positive facultative bacteria, such as Actinomyces (17%) and Streptococcus (8.2%). Of 133 strains isolated from inflamed anchor plate crevices of three subjects, 110 (83%) were anaerobic bacteria, while predominant isolates were Gram-negative rods, such as Prevotella (47%), Fusobacterium (33%) and Campylobacter (16%). On the other hand, of 146 strains isolated from healthy gingival crevices of seven subjects, 98 (67%) were facultative bacteria, while 45 (31%) were anaerobic bacteria. Predominant isolates were Gram-positive facultative bacteria, such as Actinomyces (37%) and Streptococcus (20%). CONCLUSIONS: These results suggest that the environment in crevices around titanium orthodontic anchor plates is anaerobic and supportive of anaerobic growth of bacteria, which may trigger inflammation in the tissue around the plates.  相似文献

目的:了解葡聚糖结合蛋白(CbP)A抗体对变形链球菌黏附的影响情况。方法:利用核素闪烁计数法,测定不同浓度的葡聚糖结合蛋白抗体影响两种变形链球菌对羧基磷灰石(HA)的黏附率。结果:葡聚糖结合蛋白抗体可明显抑制两种变形链球菌对羧基磷灰石的黏附率,并且随着葡聚糖结合蛋白抗体浓度的提高,抑制作用增强。结论:葡聚糖结合蛋白抗体可抑制变形链球菌对羧基磷灰石的黏附,在免疫防龋方面具有一定的意义。  相似文献

The purpose of the present study was to use terminal restriction fragment length polymorphism analysis and the 16S rRNA gene clone library to investigate the diversity of the microbiota associated with asymptomatic and symptomatic endodontic infections and to compare the bacterial community structure in these two clinical conditions. Samples were taken from asymptomatic endodontic infections associated with chronic periradicular lesions and from symptomatic infections clinically diagnosed as acute abscesses. 16S rRNA genes from DNA isolated from clinical samples were used to construct clone libraries or were subjected to terminal restriction fragment length polymorphism analysis. Sequence analysis of 186 clones revealed 42 taxa; 23 (55%) were uncultivated phylotypes, of which seven were unique to endodontic infections. Clone sequencing and terminal restriction fragment length polymorphism analysis revealed that the most commonly detected taxa were Fusobacterium nucleatum (including terminal restriction fragment types 1 and 2), Peptostreptococcus micros/Peptostreptococcus sp. oral clone AJ062/BS044/FG014, Prevotella species, Dialister species, Mogibacterium species, Lachnospiraceae oral clone 55A-34, Filifactor alocis, Megasphaera sp. oral clone CS025/BS073, and Veillonella sp. oral clone BP1-85/Veillonella dispar/V. parvula. Bacteroides-like sp. oral clone X083/Bacteroidales oral clone MCE7_20 and Dialister sp. oral clone BS016/MCE7_134 were detected only in asymptomatic teeth. On the other hand, F. nucleatum terminal restriction fragment type 2, Prevotella intermedia, Dialister pneumosintes, and some phylotypes were exclusively detected in symptomatic samples. Bacterial profiles of symptomatic endodontic infections generated by terminal restriction fragment length polymorphism analysis were clearly different from those of asymptomatic infections. Overall, the average number of terminal restriction fragments in symptomatic samples was significantly larger than in asymptomatic samples. Molecular analysis of the microbiota associated with symptomatic or asymptomatic endodontic infections indicates that the endodontic bacterial diversity is greater than previously described by culture methods and that the structure of the microbiota differ significantly between asymptomatic and symptomatic infections.  相似文献

目的:构建用于转化变形链球菌,含有F-ATP酶基因的重组质粒。方法:采用PCR方法,以变形链球菌基因组DNA为模板,扩增F-ATP酶β亚基5′末端序列,将克隆片段与载体pVA891酶切后连接,形成重组质粒,并对变形链球菌的转化作了初步分析。结果:构建的重组质粒经PCR鉴定、酶切鉴定和DNA序列测定,显示插入的目的片段序列正确。结论:变形链球菌目的片段与穿梭载体重组后能有效克隆,为通过同源重组特异突变变形链球菌染色体基因奠定基础。  相似文献

OBJECTIVE: A supernumerary cheek tooth occurs mesially to the first molar in tabby/EDA (Ta) mice affected by hypohidrotic ectodermal dysplasia. The supernumerary tooth (S) has been hypothetically homologized to the premolar, which has disappeared during mouse evolution. DESIGN: This hypothesis was tested using available morphological data on the lower cheek teeth in wild type (WT) and Ta mice. RESULTS: The presence of S is accompanied by a reduction in the mesial portion of the M(1) in mutant mice. 3D reconstructions suggest that the S in Ta homo/hemizygous embryos originates from a split off the mesial portion of the first molar (M(1)) cap. In WT embryos, two vestigial tooth primordia are transiently distinct in front of the M(1). The distal vestige has the form of a wide bud and participates during the development of the mesial portion of the M(1). This bud has been homologized with the vestigial primordium of the fourth premolar of mouse ancestors. The premolar disappearance coincided with a mesial lengthening of the M(1) during mouse evolution. The incorporation of the distal premolar vestige into the mesial part of the M(1) in WT embryos can be regarded as a repetition of the premolar disappearance during evolution. CONCLUSION:: Ontogenetic and phylogenetic data support that the S in Ta mice arises due to the segregation of the distal premolar vestige from the molar dentition and thus represents an evolutionary throwback (atavism).  相似文献

The aim of this study was to compare the detection frequencies of 25 bacterial species in subgingival and supragingival plaque of 18 untreated periodontitis subjects and 12 periodontally healthy subjects. Genomic DNA was extracted from subgingival and supragingival plaque samples, and bacterial detection was performed by polymerase chain reaction of the 16S rRNA genes. Fourteen bacteria showed no relationship with periodontitis, and 11 of these 14 species were frequently detected (≥50%) in subgingival plaque in both periodontitis and healthy subjects. Nine bacteria such as Eubacterium saphenum, Prevotella intermedia, and Treponema denticola seemed to be related to periodontitis; their detection frequencies in subgingival plaque samples were higher in periodontitis than in healthy subjects, but these differences were not statistically significant by multiple comparisons (0.002≤ P< 0.05). Two species ( Mogibacterium timidum and Porphyromonas gingivalis ) were detected significantly more frequently in subgingival plaque of periodontitis subjects than of healthy subjects ( P< 0.002), with P. gingivalis being detected only in periodontitis subjects, suggesting that these two species are closely related to periodontitis. There were no significant differences in the detection frequencies of the 25 bacteria between subgingival and supragingival plaque, suggesting that the bacterial flora of supragingival plaque reflects that of subgingival plaque.  相似文献
